<dcterms:abstract xml:lang="fr">Le tissu adipeux est un organe métabolique dynamique avec des fonctions endocriniennes, immunitaires et thermogéniques qui s'étendent bien au-delà du stockage d'énergie. Cette thèse examine trois aspects fondamentaux de la biologie du tissu adipeux : la différenciation adipocytaire, la rythmicité circadienne et le métabolisme du glycogène. Nous avons tout d'abord établi un protocole robuste pour isoler et différencier les préadipocytes blancs et bruns d'embryons de souris au jour embryonnaire 15,5 (E15.5). Cette approche répond à une lacune méthodologique critique et fournit un cadre pour étudier comment l'identité adipocytaire s'établit durant le développement périnatal, lorsque des conditions environnementales défavorables peuvent programmer une susceptibilité à vie à l'obésité, à la résistance à l'insuline et au diabète de type 2.En utilisant ce protocole de différenciation conjointement avec du tissu adipeux brun (TAB) adulte, nous avons examiné les effets de la désynchronisation circadienne chronique (DCC) sur la morphologie et la fonction du TAB. La taille des gouttelettes lipidiques dans le TAB suit des rythmes circadiens robustes déterminés par des horloges moléculaires cellule-autonomes dans les adipocytes bruns. Nous avons soumis des souris à une DCC par inversions hebdomadaires de la phase lumière-obscurité pendant 23 semaines. Bien que cette perturbation ait modifié la phase des oscillations des gouttelettes lipidiques, leur rythmicité a été maintenue. Après une resynchronisation de deux semaines, le TAB a entièrement préservé sa morphologie, son expression d'UCP1 et sa thermogenèse induite par le froid. En contraste, le brunissement du tissu adipeux blanc sous-cutané a été nettement altéré, suggérant que différents dépôts adipeux présentent des vulnérabilités distinctes à la perturbation circadienne.Nous démontrons que le glycogène sert de régulateur dynamique de la thermogenèse adaptative plutôt que de réserve énergétique passive. À travers des expositions successives au froid, le glycogène du TAB subit des cycles coordonnés de synthèse et de dégradation essentiels pour maintenir la capacité thermogénique. En utilisant des modèles murins génétiquement modifiés, nous montrons que la dégradation enzymatique du glycogène via l'enzyme débranchante AGL est indispensable pour maintenir la température corporelle sous stress métabolique, tandis que la glycophagie est dispensable. La perte de fonction d'AGL conduit à une accumulation de glycogène, une déplétion des gouttelettes lipidiques, une désorganisation mitochondriale et une altération profonde de l'intégration métabolique systémique. Ces résultats positionnent le glycogène comme un hub régulateur coordonnant le métabolisme des glucides, des lipides et des protéines dans les adipocytes thermogéniques. Cette thèse redéfinit le tissu adipeux comme un système hautement coordonné dans lequel la programmation développementale, la rythmicité circadienne et l'utilisation des substrats métaboliques convergent pour déterminer la capacité fonctionnelle. Ces résultats font progresser notre compréhension de la façon dont le tissu adipeux s'adapte aux défis environnementaux et offrent de nouvelles perspectives sur la résilience métabolique, la plasticité tissulaire et les interventions potentielles pour l'obésité et les troubles métaboliques associés.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Adipose tissue has emerged as a dynamic metabolic organ with endocrine, immune, and thermogenic functions that extend far beyond energy storage. This thesis investigates three fundamental aspects of adipose tissue biology: adipocyte differentiation, circadian rhythmicity, and glycogen metabolism.In this study, we first established a robust protocol for isolating and differentiating white and brown preadipocytes from mouse embryos at embryonic day 15.5 (E15.5). This approach addresses a critical methodological gap and provides a framework for studying how adipocyte identity is established during perinatal development, when adverse environmental conditions can program lifelong susceptibility to obesity, insulin resistance, and type 2 diabetes.Using this adipocyte differentiation protocol alongside adult BAT, we examined the effects of chronic clock desynchronization (CCD) on BAT morphology and function in mice. We found that lipid droplets (LDs) size in BAT follows robust circadian rhythms driven in part by cell-autonomous molecular clocks in brown adipocytes. To test the resilience of this circadian pattern, we subjected mice to CCD through weekly light-dark phase inversions for up to 23 weeks. Remarkably, while this perturbation altered the phase of LDs oscillations, their rhythmicity was maintained. Following a two-week resynchronization period after a CCD, BAT fully preserved its morphology, UCP1 expression, and cold-induced thermogenesis. In striking contrast, browning of subcutaneous white adipose tissue was markedly impaired by CCD, suggesting that different adipose depots exhibit distinct vulnerabilities to the same circadian perturbation. Lastly, we demonstrate that glycogen serves as a dynamic regulator of adaptive thermogenesis rather than a passive energy reserve. Through successive cold exposures, BAT glycogen undergoes coordinated synthesis and degradation cycles essential for sustaining thermogenic capacity. Using genetically modified mouse models, we show that enzymatic glycogen breakdown via the debranching enzyme AGL is indispensable for maintaining body temperature under metabolic stress, whereas glycophagy is dispensable for acute energy production. Loss of AGL function leads to glycogen accumulation, lipid droplet depletion, mitochondrial disorganization, and profound impairment of systemic metabolic integration. These findings position glycogen as a regulatory hub coordinating carbohydrate, lipid, and protein metabolism in thermogenic adipocytes, with implications for understanding glycogen storage diseases where environmental temperature may modify disease severity.In conclusion, this thesis redefines adipose tissue as a highly coordinated system in which developmental programming, circadian rhythmicity, and metabolic substrate utilization converge to determine functional capacity. The first pillar of this thesis involves establishing embryonic preadipocyte differentiation models, which enables the investigation of the mechanisms behind early programming events that influence lifelong metabolic health. The second finding demonstrate that BAT maintains functional resilience despite CCD, while WAT does not, highlighting tissue-specific regulatory networks that may be therapeutically targetable. The third parallel finding identifies glycogen as a central regulator, rather than merely a storage molecule, demonstrate how thermogenic adipocytes achieve metabolic flexibility through the coordinated use of metabolic pathways. Merged together, these findings advance our understanding of how adipose tissue adapts to environmental challenges and offer new perspectives on metabolic resilience, tissue plasticity, and potential interventions for obesity and related metabolic disorders.</dcterms:abstract>
