<dcterms:abstract xml:lang="fr">Aujourd'hui, le cancer du sien est la première cause de mortalité par cancer chez la femme dans le monde, en grande partie en raison de la survenue de métastases et de la résistance aux traitements. Ces dernières années, de nombreuses études ont mis en lumière le rôle des ARN non codants dans la progression tumorale. Parmi eux, le long ARN non codant H19 a émergé comme un acteur clé dans plusieurs types de cancer, mais ses mécanismes d'actions restent encore mal compris. Au cours de cette thèse, j'ai étudié l'implication d'H19 dans l'émergence des phénotypes agressifs du cancer du sein. J'ai montré qu'H19 module l'expression de BST2, une glycoprotéine transmembranaire impliquée dans l'adhésion et la signalisation cellulaire. De manière intéressante, mes résultats révèlent une régulation croisée entre H19 et BST2, médiée par le microARN miR-760, qui cible spécifiquement ces deux ARN. Cette régulation conjointe contribue à la prolifération, la migration, l'invasion et la tumorigénicité des cellules cancéreuses mammaires. En parallèle, nous montrons que l'expression d'H19 module également le métabolisme énergétique des cellules tumorales, favorisant la respiration mitochondriale au détriment de la glycolyse dans un modèle cellulaire tumoral. Ainsi, ces travaux mettent en évidence un nouvel axe de régulation post-transcriptionnelle H19/miR-760/BST2 dans le cancer du sein, en lien avec l'agressivité tumorale. De plus, ils soulignent également l'intérêt d'H19 comme régulateur du métabolisme énergétique dans les cellules tumorales mammaires. Ainsi, cette étude offre un nouvel éclairage sur le rôle des ARN non codants dans la tumorigenèse, et ouvre la voie à de nouvelles approches thérapeutiques ciblant ces molécules dans le cancer du sein.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Breast cancer is currently the leading cause of cancer-related death among women worldwide, largely due to the development of metastases and resistance to treatment. In recent years, numerous studies have highlighted the role of non-coding RNAs in tumor progression. Among them, the long non-coding RNA H19 has emerged as a key player in several cancer types, although its mechanisms of action remain poorly understood. In this thesis, we investigated the involvement of H19 in the acquisition of aggressive phenotypes in breast cancer. We show that H19 regulates the expression of BST2, a transmembrane glycoprotein involved in cell adhesion and tumor development. Interestingly, our results reveal a reciprocal regulation between H19 and BST2, mediated by the microRNA miR-760, which directly targets both RNAs. This combined regulation promotes proliferation, migration, invasion and tumorigenicity of breast cancer cells. In parallel, we demonstrate that H19 expression also modulates the energy metabolism of breast tumor cells. Thus, our findings highlight a novel post-transcriptional regulatory axis, H19/miR-760/BST2, linked to breast cancer aggressiveness. Moreover, they emphasize the role of H19 as a potential regulator of energy metabolism in tumor cells. This study provides new insights into the role of non-coding RNAs in tumorigenesis and paves the way for the development of therapeutic strategies targeting these molecules in breast cancer.</dcterms:abstract>
