<dcterms:abstract xml:lang="fr">Malgré les vaccins et des traitements antiviraux disponibles, les infections respiratoires virales restent une cause importante de morbidité et de mortalité à l’échelle mondiale. Les individus âgés, particulièrement sensibles aux infections respiratoires virales, présentent un taux de mortalité supérieur à celui des adultes plus jeunes, ainsi qu’une période de récupération plus longue. Nous avons émis l’hypothèse que la sénescence des cellules pulmonaires, qu’elle soit préexistante (liée à l’âge) ou induite par le virus, joue un rôle dans la gravité des infections respiratoires virales, tant au niveau de la phase aiguë que de la phase chronique. La sénescence cellulaire est un processus biologique qui résulte d’un stress cellulaire, et qui aboutit à un arrêt de la division cellulaire, à une résistance à l’apoptose, ainsi qu’à la synthèse de facteurs inflammatoires ou de facteurs liés à la réparation tissulaire. À l’aide de modèles précliniques d’infection par les virus Influenza A (IAV) et SARS-CoV-2 (l’agent du COVID-19), nous démontrons que ces virus entraînent une accumulation de cellules sénescentes dans les poumons, même après l’élimination complète du virus. Nous avons caractérisé les types cellulaires devenus sénescents, analysé la présence de virus dans ces cellules, et examiné le rôle des cellules sénescentes dans la pathologie pulmonaire. Nos travaux révèlent que, selon le modèle utilisé et l’âge des animaux, la sénescence cellulaire peut affecter la réplication virale, l’intensité de l’inflammation pulmonaire ainsi que les processus de réparation, notamment la régénération de l’épithélium bronchique. Ainsi, l’élimination des cellules sénescentes, par des approches génétiques (souris p16-ATTAC) et/ou pharmacologiques (Navitoclax), atténue l’inflammation pulmonaire et systémique, et limite les séquelles à long terme de l’infection. Ces effets protecteurs sont particulièrement marqués chez les animaux âgés, pour qui la sénescence préexistante exacerbe la sévérité de la maladie et retarde la régénération tissulaire. Ce projet met ainsi en évidence le rôle de la sénescence cellulaire pulmonaire dans la pathogenèse des infections respiratoires virales, à court et à long terme. Il ouvre la voie à une approche thérapeutique reposant sur l’utilisation de sénolytiques, en vue d’atténuer la sévérité des formes aiguës et de prévenir les complications chroniques associées à ces infections.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Despite the availability of vaccines and antiviral treatments, viral respiratory infections remain a major cause of morbidity and mortality worldwide. Elderly individuals, who are particularly vulnerable to viral respiratory infections, show higher mortality rates than younger adults, as well as longer recovery periods. We hypothesized that the senescence of lung cells, whether preexisting (age-related) or virus-induced, plays a role in the severity of viral respiratory infections, both during the acute and chronic phases. Cellular senescence is a biological process resulting from cellular stress, leading to cell cycle arrest, resistance to apoptosis, and the production of inflammatory or tissue-repair–related factors.Using preclinical models of infection with Influenza A virus (IAV) and SARS-CoV-2 (the causative agent of COVID-19), we demonstrate that these viruses induce an accumulation of senescent cells in the lungs, even after complete viral clearance. We characterized the cell types that became senescent, analyzed the presence of virus within these cells, and examined the role of senescent cells in lung pathology. Our studies reveal that, depending on the model used and the age of the animals, cellular senescence can affect viral replication, the intensity of pulmonary inflammation, and repair processes, including epithelial regeneration. Accordingly, the elimination of senescent cells through genetic (p16-ATTAC mice) and/or pharmacological (navitoclax) approaches reduces pulmonary and systemic inflammation and limits the long-term sequelae of infection. These protective effects are particularly pronounced in aged animals, in which preexisting senescence exacerbates disease severity and delays tissue regeneration. This project therefore highlights the role of pulmonary cellular senescence in the short- and long-term pathogenesis of viral respiratory infections. It opens the way to a therapeutic approach based on the use of senolytic agents to reduce the severity of acute disease and to prevent the chronic complications associated with these infections.</dcterms:abstract>
