<dcterms:abstract xml:lang="fr">Ce travail de thèse s'est concentré sur l'étude d'ACSL4, une enzyme clé du métabolisme lipidique récemment identifiée comme un acteur central de la ferroptose, une forme de mort cellulaire régulée impliquée dans les maladies neurodégénératives. Malgré l'émergence récente de quelques inhibiteurs d'ACSL4, ces molécules présentent quelques faiblesses et on note une absence de véritable démarche de chimie médicinale. Ce projet de thèse s'est donc donné pour ambition de développer de nouveaux inhibiteurs puissants et sélectifs d'ACSL4 en tant qu'outils anti-ferroptotiques. Le premier chapitre, fondé sur une stratégie SOSA (Selective Optimization of Side Activities) à partir de la rosiglitazone, a permis le développement d'une nouvelle série d'inhibiteurs puissants d'ACSL4. Parmi eux, le composé 12 (IC50 = 105 nM) protège les neurones dopaminergiques de la ferroptose et réduit la peroxydation lipidique. Des études structurales ont révélé le mode de liaison de ces inhibiteurs dans la poche à acides gras, ce qui a permis la construction d'un modèle tridimensionnel prédictif d'ACSL4 robuste, ouvrant la voie à des optimisations.Le second chapitre repose sur une approche FBDD (Fragment-Based Drug Design) ayant permis d'identifier 31 fragments actifs sur ACSL4, dont un dérivé benzofuranique optimisé en composé 49 (IC50 = 49 nM). Bien que très affin, sa faible solubilité a motivé une optimisation structurale, notamment par l'introduction de contraintes stériques, menant au composé 74, doté d'une solubilité significativement améliorée et d'une bonne affinité et sélectivité. En résumé, ce travail de thèse a conduit à la découverte d'inhibiteurs puissants et sélectifs d'ACSL4, enrichissant le pipeline existant tout en apportant des avancées structurales. Les deux séries développées comptent désormais de nombreux analogues actifs, offrant une base solide pour l'exploration des relations structure-activité (RSA). Plusieurs composés présentent des activités enzymatiques de l'ordre du nanomolaire, corrélées à une efficacité cellulaire satisfaisante, ce qui constitue une preuve de concept forte et ouvre la porte à la validation in vivo des candidats les plus prometteurs.</dcterms:abstract>
<dcterms:abstract xml:lang="en">This PhD work focused on the study of ACSL4, a key enzyme in lipid metabolism recently identified as a central player in ferroptosis, a regulated form of cell death implicated in neurodegenerative diseases. Despite the recent emergence of a few ACSL4 inhibitors, these molecules suffer from several limitations, and a true medicinal chemistry effort is still lacking. The aim of this thesis was therefore to develop novel, potent, and selective ACSL4 inhibitors as anti-ferroptotic tools. The first chapter is based on a SOSA (Selective Optimization of Side Activities) strategy starting from rosiglitazone, which led to the development of a new series of potent ACSL4 inhibitors. Among them, compound 12 (IC?? = 105 nM) protects dopaminergic neurons from ferroptosis and reduces lipid peroxidation. Structural studies revealed the binding mode of these inhibitors within the fatty acid pocket, allowing the construction of a robust and predictive three-dimensional model of ACSL4, paving the way for further optimization. The second chapter relies on a Fragment-Based Drug Design (FBDD) approach that enabled the identification of 31 active fragments targeting ACSL4, including a benzofuran derivative optimized into compound 49 (IC?? = 49 nM). Despite its high affinity, its poor solubility prompted structural optimization, notably through the introduction of steric constraints, leading to compound 74, which combines significantly improved solubility with good affinity and selectivity.In summary, this thesis led to the discovery of potent and selective ACSL4 inhibitors, enriching the current inhibitor pipeline while providing key structural insights. The two developed series now include numerous active analogues, forming a solid foundation for the exploration of structure-activity relationships (SAR). Several compounds exhibit nanomolar enzymatic activity correlated with robust cellular efficacy, constituting a strong proof of concept and opening the way to in vivo validation of the most promising candidates.</dcterms:abstract>
