<dcterms:abstract xml:lang="fr">Les cannabinoïdes, au sens large de ce terme (endocannabinoïdes, cannabis, cannabidiol, cannabinoïdes de synthèse ou d’hémisynthèse), font partie des sujets d’actualité pour des raisons scientifiques, sociétales et médiatiques. Cependant, et malgré de nombreuses recherches dans le domaine médical, plusieurs aspects pharmacologiques et toxicologiques de ces substances demeurent mal connus. La complexité de recherche analytique des molécules agissant sur les récepteurs cannabinoïdes réside d’une part, dans la multitude et la variété des cannabinoïdes endogènes (endocannabinoïdes) et exogènes (phytocannabinoïdes, cannabinoïdes de synthèse et d’hémisynthèse), et d’autre part dans leurs particularités physicochimiques, des concentrations circulantes généralement très faibles dans les matrices biologiques et un métabolisme parfois intense. Compte tenu de ces difficultés, la capacité d’identifier et de quantifier ces molécules présente de nombreux écueils sur les plans pré-analytique, analytique et post-analytique. L’objectif de ma thèse était d’appréhender ces difficultés et d’étudier les implications des endocannabinoïdes, et des cannabinoïdes en général, dans les domaines de la toxicologie biologique et médico-légale à travers le développement d’outils analytiques permettant leurs mises en évidence dans les différentes matrices biologiques.A cet effet, trois méthodes analytiques de quantification d’endocannabinoïdes et de substances endocannabinoïdes-like dans trois matrices biologiques différentes (plasma, cheveux et spot de sang séché - DBS), ainsi qu’un outil d’exploration de métabolisme in silico des cannabinoïdes de synthèse, ont été développées. Ces outils ont été mis en oeuvre dans 3 projets de recherche et ont permis (i) d’étudier la relation entre cannabinoïdes sériques et insulinorésistance chez des personnes coinfectées VIH-VHC et d’explorer les effets (de la guérison VHC et de la prise de cannabinoïdes) sur les endocannabinoïdes, (ii) d’étudier l’évolution des endocannabinoïdes dans le sang en période post-mortem, et (iii) d’étudier le métabolisme de 13 « nouveaux » cannabinoïdes de synthèse, pour certains, non décrit précédemment dans la littérature, circulant en 2023 sur l’île de Mayotte, France.Au total, ce travail réalisé au cours de ces trois années de thèse m’a permis d’élaborer des outils analytiques de détection et quantification d’endocannabinoïdes et de cannabinoïdes exogènes dans différentes matrices (et avec des méthodes alternatives de prélèvement sanguin) ; outils complétés de procédures in silico de prédiction à la fois du métabolisme et des paramètres analytiques de biomarqueurs (métabolites) d’exposition qui pourront être appliquées à un périmètre élargi en toxicologie biologique et médicolégale.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Cannabinoids, in the broad sense of this word (endocannabinoids, cannabis, cannabidiol, synthetic or semi-synthetic cannabinoids), are a hot topic for scientific, societal and media reasons. However, despite extensive medical researchs, many pharmacological and toxicological aspects of these substances remain unclear. The complexity of analytical research on molecules that interact with cannabinoid receptors lies, on the one hand, in the multitude and diversity of endogenous (endocannabinoids) and exogenous (phytocannabinoids, synthetic and semi-synthetic cannabinoids) compounds and, on the other hand, in their physicochemical characteristics, their generally very low circulating concentrations in biological matrices and their sometimes-intense metabolism. Given these difficulties, the ability to identify and quantify these molecules presents many pitfalls at the pre-analytical, analytical and post-analytical stages. The aim of my research was to understand these difficulties and explore the implications of endocannabinoids and cannabinoids in general terms, in the fields of biological and forensic toxicology through the development of analytical tools for their detection in different biological matrices. In this way, three analytical methods were developed in order to quantify endocannabinoids and endocannabinoid-like substances in three different biological matrices (plasma, hair and dried blood spots - DBS), as well as a tool to in silico explore the metabolism of synthetic cannabinoids. These tools were implemented in three research projects and allowed (i) to study the relationship between plasma cannabinoids and insulin resistance in people co-infected with HIV and HCV and to explore the effects (of HCV cure and cannabinoid use) on endocannabinoids, (ii) study the evolution of endocannabinoids in the blood during the post-mortem period, and (iii) study the metabolism of 13 ‘new’ synthetic cannabinoids, some of which have never been described in the literature, circulating in 2023 on the island of Mayotte, France. Overall, this work has enabled me to develop analytical tools for the detection and quantification of endocannabinoids and exogenous cannabinoids in different matrices (and using alternative blood sampling methods). These tools are complemented by in silico procedures for predicting both the metabolism and analytical parameters of exposure biomarkers (metabolites), which can be applied to a wider range of applications in biological and forensic toxicology.</dcterms:abstract>
