<dcterms:abstract xml:lang="fr">Les candidoses invasives représentent les infections fongiques les plus fréquemment rencontrées à l'échelle mondiale, touchant près de 1,56 million de personnes par an et provoquant environ un million de décès. En réponse à cette menace croissante, l'Organisation Mondiale de la Santé (OMS) a publié fin 2022 une première liste de pathogènes fongiques prioritaires, dans le but de guider la recherche et d'encourager le développement de nouveaux outils diagnostiques et thérapeutiques. Le genre Candida y figure en bonne place, avec Candida albicans et Candida auris classés comme pathogènes critiques, C. glabrata, C. parapsilosis et C. tropicalis comme hautement prioritaires, et C. krusei comme priorité moyenne.Le diagnostic souvent tardif et la montée de résistances aux quatre classes actuelles d'antifongiques (polyènes, azolés, échinocandines, dérivés pyrimidines) limitent fortement les options thérapeutiques disponibles en milieu hospitalier. Identifier de nouvelles cibles fongiques devient crucial, en particulier pour contrer les souches émergentes multirésistantes comme C. auris. Dans ce contexte, le développement d'inhibiteurs de l'enzyme tréhalose-6-phosphate-phosphatase TPS2 a débuté au sein du laboratoire et a permis le développement des hydrazones comme inhibiteurs potentiels. Notre travail s'est ensuite concentré sur une nouvelle famille chimique identifiée lors de ces travaux et qui s'est démarquée pour son activité antifongique in vitro. Une étude de pharmacomodulation a été menée afin d'explorer les relations structure-activité et d'optimiser cette famille. Nous avons déterminé les concentrations minimales inhibitrices (CMI) sur plusieurs souches cliniques de Candida, et évalué l'effet des composés sur la croissance, le développement morphologique et la formation de biofilm de C. albicans, notamment par imagerie. L'innocuité des molécules a également été testée sur une lignée de cellules humaines. En parallèle, nous avons entrepris l'identification des cibles moléculaires de ces composés à l'aide de méthodes innovantes, dites "label-free", permettant d'étudier les interactions protéine-ligand sans marquage. Deux approches ont été utilisées : la précipitation de protéines induite par solvant (SIP) et le profilage thermique du protéome (TPP). Ces techniques ont permis d'identifier plusieurs cibles potentielles, dont quatre sont absentes chez les vertébrés, renforçant leur intérêt thérapeutique. Pour valider ces cibles, des souches de C. albicans surexprimant ou délétant les gènes correspondants ont été générées par la technologie CRISPR-Cas9. L'impact de ces modifications sur différents aspects de la physiologie fongique a été étudié : croissance, morphogénèse, ainsi que l'interaction avec des macrophages humains. Enfin, un modèle in vivo basé sur la larve de Galleria mellonella a été développé au laboratoire pour évaluer à la fois la toxicité des composés et leur efficacité contre la candidose invasive. Ce modèle, largement reconnu dans la littérature, a permis de débuter l'évaluation préclinique des molécules les plus prometteuses. Ce travail constitue une avancée dans la recherche de nouvelles stratégies antifongiques face aux pathogènes émergents multirésistants, et apporte des éléments originaux pour l'identification de nouvelles cibles thérapeutiques fongiques.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Invasive candidiasis is the most frequently encountered fungal infection worldwide, affecting nearly 1.56 million people annually and causing approximately one million deaths. In response to this growing threat, the World Health Organization (WHO) published in late 2022 its first list of priority fungal pathogens, aiming to guide research efforts and promote the development of new diagnostic and therapeutic tools. The Candida genus features prominently in this list, with Candida albicans and Candida auris classified as critical priority pathogens, C. glabrata, C. parapsilosis, and C. tropicalis as high-priority, and C. krusei as medium-priority. Frequent delays in diagnosis and the increasing resistance to the four currently available classes of antifungal drugs (polyenes, azoles, echinocandins, and pyrimidine derivatives) significantly limit treatment options in hospital settings. Identifying new fungal targets has therefore become essential, particularly to treat emerging multidrug-resistant strains such as C. auris.In this context, the development of inhibitors targeting the trehalose-6-phosphate phosphatase enzyme TPS2 was initiated in our laboratory, leading to the identification of hydrazones as potential inhibitors. Building on this work, our focus shifted to a new chemical family identified during this project, which demonstrated notable in vitro antifungal activity. A pharmacomodulation study was conducted to explore structure-activity relationships and optimize this chemical scaffold. We determined the minimum inhibitory concentrations (MICs) for various clinically relevant Candida strains and assessed the effects of these compounds on yeast growth, morphogenesis, and C. albicans biofilm formation, notably through imaging techniques. The safety of the compounds was also evaluated on a human cell line.In parallel, we pursued the identification of molecular targets of these compounds using innovative, label-free techniques for studying protein-ligand interactions. Two complementary approaches were employed: solvent-induced protein precipitation (SIP) and thermal proteome profiling (TPP). These methods revealed several potential targets, four of which are absent in vertebrates, thereby increasing their therapeutic relevance. To validate these targets, C. albicans strains overexpressing or deleting the corresponding genes were generated using CRISPR-Cas9 technology. The impact of these genetic modifications on fungal physiology was assessed, including growth, morphogenesis, and interactions with human macrophages. Finally, an in vivo model based on Galleria mellonella larvae was developed in the laboratory to assess both compound toxicity and antifungal efficacy in the context of invasive candidiasis. This model, widely recognized in the scientific literature, enabled the initiation of preclinical evaluation of the most promising molecules. This work represents a step forward in the search for novel antifungal strategies against emerging multidrug-resistant pathogens and contributes original insights into the identification of new fungal therapeutic targets.</dcterms:abstract>
