<dcterms:abstract xml:lang="fr">La répression Polycomb exerce une régulation transcriptionnelle par deux complexes protéiques distincts, les complexes PRC1 et PRC2. Ce contrôle épigénétique est médié par la marque épigénétique H2AK119ub1 déposée par PRC1 et la marque H3K27me3 catalysée par PRC2. Découvert d'abord chez la drosophile, les deux complexes ont été conservés et se sont diversifiés au cours de l'évolution. Une multiplication du nombre de protéines de la famille Polycomb est observée chez les vertébrés. Ces protéines peuvent moduler l'action des complexes et participer au recrutement sur des cibles génomiques spécifiques. La répression Polycomb applique une régulation négative sur de nombreux gènes impliqués dans des grands processus biologiques mais aussi dans l'apparition et la progression de tumeurs. Cette étude s'intéresse donc au rôle de la répression Polycomb dans le développement et dans le cancer, sur le modèle poisson zèbre.Les gènes cbx sont conservés chez les vertébrés et plus particulièrement chez les téléostéens. Les gènes cbx sont tous exprimés au cours du développement et au stade adulte chez le poisson zèbre, avec des profil d'expression similaire, même si on dénote quelques différences. Si il y a donc des redondances fonctionnelles, il semble exister des spécificités fonctionnelles. L'histoire évolutive de Ezh1 et Ezh2 a été étudiée par des analyses in silico, et Ezh1 semble être le fruit d'une duplication de Ezh2. L'expression de ezh2 et ezh1 est inversement proportionnelle au cours du développement. Une étude fonctionnelle de Ezh2 et Ezh1 révèle que la compensation de la perte de fonction de Ezh2 par Ezh1 est minime. L'activité totale du complexe PRC2 dans le développement du poisson zèbre a été analysé par la génération d'un nouveau mutant eed. La mutation est létale autour de 11 jpf en raison d'altérations importantes du système digestif. La perte de la marque H3K27me3 altère la différenciation des cellules neurales. Les larves eed-/- reproduisent le paysage épigénétique observé dans certains cancers dont l'activité PRC2 est inhibée.. La lignée mutante pour eed est donc un modèle pertinent pour l'étude de cancers impliquants la répression Polycomb, et notamment les DIPGs.Enfin, deux revues de littérature ont été écrites, afin de rassembler les connaissances actuelles sur les expérimentations de transplantations de cellules cancéreuses chez le poisson zèbre, ainsi que l'utilisation de l'ingénierie génétique dans la modélisation de cancers sur le modèle poisson zèbre.</dcterms:abstract>
<dcterms:abstract xml:lang="en">The Polycomb repression exerts transcriptional control by two distinct complexes, PRC1 AND PRC2. This epigenetic control is ensured by H2AK119ub1 and H3K27me3 deposition. The polycomb repression was firstly discovered in Drosophila. The analyses of genic and proteic sequences revealed that the Polycomb protein family has been conserved and diversified through evolution. A high number of Polycomb family proteins is present in vertebrates. These proteins can modulate the activity the action of the complexes and take part to their recruitment to specific genomic loci. The Polycomb repression negatively regulates many genes involved in major biological processes, but also in the initiation and progression of cancers. Therefore, this study focuses on the role of Polycomb repression in development and in cancer, based on the zebrafish model.The cbx genes are conserved in vertebrates and more particularly in teleosts. The cbx genes are all expressed during development and in different adult organs. The expression patterns are very similar between the orthologues, although there are some differences. These results suggest the existence of functional specificity. The evolutionary history of Ezh1 and Ezh2 has been studied by in silco analyses. Ezh1 appeared to be the result of an Ezh2 duplication. The expression of ezh2 and ezh1 is inversely proportional during development. A functional study of Ezh2 and Ezh1 reveals that the compensation for the loss of function of Ezh2 by Ezh1 is slight. The total activity of the PRC2 complex in the zebrafish development was analyzed thanks to the generation of a new eed mutant line. The mutation is lethal around 11 dpf due to significant alterations of digestive organs. Loss of the H3K27me mark impairs neural cell differentiation. The eed-/- larvae reproduce the epigenetic landscape observed in tumors with an inhibition of PRC2 activity. The mutant line for eed is therefore a relevant model for the study of cancer involving the Polycomb repression.Finally, two reviews were written in order to bring together current knowledge on cancer cell transplantation experiments, as well as the use of genetic engineering in the modeling of cancers on the zebrafish model.</dcterms:abstract>
