<dcterms:abstract xml:lang="en">The neonatal period refers to an intense intestinal developmental phase during which the intestinal barrier is established. Numerous studies highlight the critical role of the early life nutritional environment in the gut development and future health. Postnatal growth restriction (PNGR) in mice is associated with delayed intestinal maturation and increased susceptibility to colitis in adulthood. breast milk has a protective role in the occurrence of digestive pathologies. These beneficial effects are attributed to its richness in bioactive factors, such as hormones or lactoferrin, an abundant milk protein with antimicrobial, anti-inflammatory and prebiotic activities. Newborns with PNGR are dependent on human milk, whose sterilization is ensured in the human milk bank by Holder Pasteurization (HoP), however leading to the loss of many bioactive factors. Thus, while few studies focus on the effects of milk bioactive factors in preventing the deleterious consequences of altered intestinal maturation, the search for alternative sterilizing treatments to HoP to preserve the bioactive qualities of milk, such as the High Hydrostatic Pressure (HHP) technique seems essential. In this context, the objectives of this work were: 1) to evaluate a new method of sterilization of human milk (HHP) on its bioactive factors, 2) to characterize the impact of a bioactive factor of breast milk, lactoferrin, on intestinal maturation. The induction of PNGR, by increasing the number of pups per litter, led at weaning to a delay in intestinal maturation, associating reduction in the depth of the crypts and overexpression of lactase at the ileal level, as well as enrichment of the intestinal microbiota in Lactobacillales. We also showed an alteration of the intestinal transcriptome by PNGR with overexpression of Per2, Per3, Tef and Usp2, and underexpression of Bmal1 and Rev-erb?, which are circadian clock genes regulating rhythmic biological processes. Supplementation with bLf during lactation did not prevent all of these variations but reduced the richness of the microbiota of control mice at weaning while modulating its composition in Bacteroidales, Gammaproteobacteria and Clostridia. Finally, the induction of acute colitis after weaning was not amplified by PNGR nor prevented by bLf supplementation. Regarding the preservation of milk bioactive factors, we showed that the HHP technique better preserved the concentrations of some metabolic hormones, such as insulin, leptin, cortisol, apelin, GLP-1 and nesfatin-1, compared to the HoP. Adiponectin was however degraded by HHP. Also, treatment with HHP allowed better preservation of lactoferrin, compared to HoP. Lysozyme, another milk antimicrobial peptide, was unaffected by both treatments. The evaluation of the antimicrobial activity of treated milks on bacterial growth over time showed a strong bacteriostatic effect against Escherichia coli as well as a greater bactericidal effect against Streptococcus agalactiae for HHP, compared to HoP.Our work emphasizes the importance of adequate intestinal maturation during the early postnatal period and reinforces the interest of bioactive factors in breast milk for their unique metabolic regulating, prebiotic or antimicrobial properties, and thus, the importance of their preservation in the context of sterilization within human milk banks.</dcterms:abstract>
