<dcterms:abstract xml:lang="fr">Le long ARN non codant H19 (lncRNA) est surexprimé dans 70% des cancers du sein et favorise les propriétés tumorigènes des cellules cancéreuses mammaires. De plus, H19 est le précurseur du miR-675, impliqué dans la tumorigenèse mammaire. L'objectif de ce projet de thèse est de décrypter, dans les cellules cancéreuses mammaires, le rôle et les mécanismes d'action du lncRNA H19 ainsi que les parts relatives de l'ARNm et du miR-675 sur les phénotypes observés lors de l'émergence des métastases.Afin de déterminer l'implication et la contribution relative de H19 et du miR-675 dans l'augmentation du potentiel métastatique du cancer du sein, nous avons discriminé les phénotypes du processus métastatique associés à l'expression du miR-675 et ceux dus à l'ARNm lui-même. Mes travaux de thèse montrent que le lncRNA H19 et le miR-675 augmentent tous deux la migration cellulaire ainsi que l'invasion des cellules cancéreuses mammaires in vitro et in vivo. Nous démontrons, pour la première fois que le miR-675 comme H19, augmentent les propriétés souches des cellules cancéreuses du sein. D'autre part, H19 et le miR-675 ont des actions séquentielles. En effet, in vitro, H19 semble induire l'EMT alors que le miR-675 semble induire la MET.Dans le but d'identifier des déterminants moléculaires régulés par H19 lors de l'émergence métastatique, nous avons réalisé une étude transcriptomique. D'après ces résultats, le gène BST2 est surexprimé dans les cellules surexprimant stablement H19. Nous mettons en évidence par des expériences de perte et de gain de fonction qu'il existe une régulation post-transcriptionnelle entre H19 et BST2. De plus, j'ai montré que le miR-675 régule négativement BST2. Enfin, comme H19, BST2 augmente la prolifération, la migration et l'invasion cellulaire dans nos cellules cancéreuses mammaires.Enfin, mes travaux de thèse mettent en évidence que H19 séquestre p53 dans le cytoplasme des cellules cancéreuses mammaires, ce qui entraine une dégradation de p53 ne pouvant plus être transloqué dans le noyau cellulaire. De plus, nous montrons que H19 interagit avec p53 et avec MDM2 pour former un complexe tripartite. De par cette interaction, H19 réprime l'activité transcriptionnelle de p53 et empêche l'arrêt du cycle cellulaire, l'induction de l'apoptose et la sénescence des cellules après dommages à l'ADN.Pour conclure, mes travaux de thèse mettent en évidence de nouveaux mécanismes impliquant H19 et son miR-675 dans la progression tumorale mammaire.</dcterms:abstract>
<dcterms:abstract xml:lang="en">The long non-coding RNA H19 (lncRNA) is overexpressed in 70% of breast cancer and promotes the tumorigenic properties of breast cancer cells. Furthermore, H19 is the precursor of miR-675, which is involved in breast tumorigenesis. The aim of this thesis project is to decipher, in breast cancer cells, the role and mechanisms of action of H19 as well as the relative contribution of mRNA and miR-675 on the phenotypes observed during the emergence of metastasis.To determine the involvement and relative contribution of H19 and miR-675 in increasing the metastatic potential of breast cancer, we discriminated between phenotypes of the metastatic process associated with miR-675 expression and those due to mRNA itself. My work shows that both lncRNA H19 and miR-675 increase cell migration and invasion of breast cancer cells in vitro and in vivo. We demonstrate, for the first time, that miR-675, like H19, increase the stemness of breast cancer cells. Moreover, H19 and miR-675 have sequential actions. Indeed, in vitro, H19 seems to induce EMT while miR-675 seems to induce MET.In order to identify molecular determinants regulated by H19 during metastatic emergence, we performed a transcriptomic analysis. According to these results, the BST2 gene is overexpressed in cells stably overexpressing H19. We demonstrate by loss- and gain-of-function experiments that there is a post-transcriptional regulation between H19 and BST2. Furthermore, I have shown that miR-675 regulates negatively BST2. Finally, like H19, BST2 increases cell proliferation, migration and invasion in our breast cancer cells.Finally, my thesis work shows that H19 sequesters p53 in the cytoplasm of breast cancer cells, which leads to a degradation of p53 that can't be translocated into the cell nucleus. Moreover, we show that H19 interacts with p53 and with MDM2 to form a tripartite complex. Through this interaction, H19 represses the transcriptional activity of p53 and prevents cell cycle arrest, induction of apoptosis and senescence of cells after DNA damages.In conclusion, my work highlights new mechanisms involving H19 and its miR-675 in breast tumor progression.</dcterms:abstract>
