<dcterms:abstract xml:lang="fr">Les éléments traces métalliques (ETM) comme le plomb, le cadmium, et dans une moindre mesure l’aluminium, sont toxiques et représentent un problème majeur de santé publique. Ils peuvent être dangereux pour la santé quelle que soit leur porte d’entrée dans l’organisme (i.e. ingestion via l’alimentation ou l’eau de boisson, inhalation). De nombreuses bactéries sont capables d’interagir avec ces ETM, pour en réduire la biodisponibilité. Les bactéries d’origine intestinale et/ou alimentaires sont des candidates particulièrement intéressantes pour la bioremédiation in vivo, étant pour la plupart inoffensives pour l’humain. Dans ce travail de thèse, nous avons évalué les capacités de fixation de 225 souches bactériennes vis-à-vis du Pb(II), Cd(II), et al(III), les dosages d’ETM ayant été réalisés par ICP-MS (Inductively Coupled Plasma – Mass Spectrometer). Des comparaisons intra- et inter-espèces ont été réalisés au sein du phylum des Firmicutes (principalement des bactéries lactiques, dont des Lactobacillus, quelques Lactococcus, Pediococcus et Carnobacterium), des Actinobacteria et des Proteobacteria. Des expériences de chélation sur des mélanges de plomb et de cadmium ont également été réalisées. Des bons candidats ont ainsi d’abord été identifiés in vitro, puis évalués in vivo chez la souris. Au cours de ce travail, un effet potentialisateur du pouvoir chélateur du Tween 80, étendu à d’autres détergents, a été mis en évidence chez les bactéries alimentaires du genre Propionibacterium pour la chélation du plomb. Des expérimentations complémentaires visant à mieux comprendre les mécanismes impliqués dans ce cas particulier ont été réalisées, dont des variations des conditions de chélation ainsi que des approches par microscopie électronique et à force atomique.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Toxic metals such as lead, cadmium, and to a lesser extent, aluminium are detrimental to health and represents a major public problem. They are health threatening whenever they are ingested by aliments or water or inhaled). Some bacteria are able to reduce the bioavailability of toxic metals by interacting with them. Intestinal and alimentary bacteria represent promising candidates for in vivo bioremediation approaches, some of these bacteria already have the “generally recognized status” status. During this thesis, we used inductively coupled plasma mass spectrometry (ICP-MS) to investigate the ability of 225 bacteria strains to remove Pb(II), Cd(II), and Al(III). Intra- and interspecies comparisons were performed among the Firmicutes (mostly lactic acid bacteria, including Lactobacillus, some Lactococcus, Pediococcus and Carnobacterium), Actinobacteria, and Proteobacteria. The capacity to remove of a mixture of lead and cadmium was also investigated. Best candidates were first identified in vitro, and were challenged in vivo in a mouse model of heavy-metal exposition. A strong boosting effect of Tween 80 on the removal of lead by Propionibacterium was observed and further extended to other detergents. In line, we investigated this observation to better understand and visualize the underlying mechanisms, using variations in the chelation conditions and distinct microscopy approaches.</dcterms:abstract>
