107 isolats bactériens, à Gram positif et catalase négative, appartenant aux groupe des bactéries lactiques, ont été isolés à partir d'échantillons de méconium de six nouveaux-nés à l'hôpital de Roubaix, dans le nord de la France. Ces bactéries ont été identifiés par spectrométrie de masse MALDI-TOF comme étant des Enterococcus faecalis. Cette identification a été confirmée par séquençage du gène 16S. Six isolats E. faecalis 14, E. faecalis 28, E. faecalis 90, E. faecalis 97, E. faecalis 101 et E. faecalis 93, montraient une activité anti-Gram négatif et Gram positif, due à la production d'acide lactique et de bactériocines. Les bactériocines produites par E. faecalis 28 et E. faecalis 93, appelées entérocines DD28 et DD93, ont été purifiés par RP-HPLC et présentent une masse moléculaire de 5205,21 et 5204,89 Da respectivement. La combinaison des entérocines DD28 et DD93, avec l’erytromycine, a montré un effet synergique sur une souche de Staphylococcus aureus résistante a la méticiline (SARM) en culture planctonique ou sous forme de biofilms. Par ailleurs nous avons également montré que la combinaison de la ticarcilline et de la céfotaxime avec la colistine ont une activité antibactérienne forte contre les souches d’E. coli producteurs de BLSE. En outre, l'étude de l’entérocine DD14, une autre bactériocine produite par E. faecalis 14, et de la nisine en combinaison avec la colistine contre des souches d'E. coli d'origine porcine sous forme planctoniques et après formation de biofilm, a montré une activité antimicrobienne remarquable contre ces souches.

107 bacterial isolates, assumed as lactic acid bacteria, were isolated from samples of meconium of six donors at Roubaix hospital, in the north of France. All these bacterial isolates were identified by MALDI-TOF mass spectrometry as Enterococcus faecalis. However, only six isolates among which E. faecalis 14, E. faecalis 28, E. faecalis 90, E. faecalis 97 and E. faecalis 101 and E. faecalis 93 were active against Gram negative bacteria (GNB) and Gram positive bacteria (GPB), through production of lactic acid and bacteriocin like inhibitory substances (BLIS). These antagonistic isolates were then fully identified by sequencing the 16S gene (16S rDNA). Bacteriocins produced by E. faecalis 28 and E. faecalis 93, called enterocin DD28 and DD93, were purified by RP-HPLC. These enterocins appeared to have a molecular mass, determined by mass spectrometry, of 5205.21 and 5204.89 Da respectively. Combination of the enterocin DD28 and DD93, with erythromycin showed a synergetic effect on Methicillin Resistant Staphylococcus aureus strain on planktonic and biofilm forming cultures. Otherwise combination of ticarcillin and cefotaxime with colistin, another antimicrobial compound, showed strongest anti-bacterial activity against ESBL producing E. coli. Moreover, the study about effect of enterocin DD14, bacteriocin produced from E. faecalis 14, and nisin in combination with colistin against planktonic and biofilm formation of Escherichia coli strains, from swine origin, showed remarkable antimicrobial activity against these strains.
