<dcterms:abstract xml:lang="fr">Les glucanes périplasmiques osmorégulés (OPG) sont des constituants du périplasme de l’enveloppe de la plupart des bactéries à Gram négatif dont la concentration augmente en réponse à la baisse d’osmolarité du milieu. Les OPG sont d’importants facteurs de virulence chez les bactéries pathogènes pour l’animal ou pour les végétaux puisque leur absence conduit à une forte diminution, voire à une perte totale de la virulence. Notre modèle d’étude, Dickeya dadantii, est une entérobactérie phytopathogène responsable de la formation d’une pourriture molle sur un large spectre de plantes hôtes. L’absence d’OPG chez un mutant opgG ou opgH provoque un phénotype pleïotrope incluant une perte totale de la virulence. L’objectif de cette thèse est de tenter de mieux comprendre le rôle des OPG au sein du périplasme. Une mutation suppressive du phénotype Opg- localisée dans le gène rcsC a été isolée. Cette mutation rcsC2 conduit à une baisse de phosphorylation de RcsB due à l’augmentation de l’activité phosphatase de RcsC. Elle restaure de nombreux phénotypes y compris la virulence sur tubercule de pomme de terre mais pas sur feuille d’endive. La protéine RcsC fait partie du système à deux composants RcsCD-RcsB. ’ensemble des phénotypes observés indiquent que l’absence d’OPG active le système RcsCD-RcsB. L’opéron opgGH a été placé sous le contrôle d’un promoteur inductible par l’arabinose, ce qui nous a permis de moduler finement la quantité d’OPG produite en fonction de la concentration d’arabinose présente dans le milieu. Nous avons ainsi pu montrer que la quantité d’OPG module l’activité du système RcsCD-RcsB. De plus, nous avons pu identifier d’autres gènes régulés par les OPG. Les résultats suggèrent que certains de ces gènes pourraient être régulés par des systèmes à deux composants différents de RcsCD-RcsB. Il semble donc que les OPG interviennent dans la signalisation cellulaire par une ou plusieurs interactions à déterminer avec différents systèmes à deux composants.</dcterms:abstract>
<dcterms:abstract xml:lang="en">The Osmoregulated Periplasmic Glucans (OPG) are general periplasmic components of the envelope of most Gram negative bacteria. Their concentration increases as the osmolarity of the medium decreases. OPG are important virulence factors in plant or animal pathogenic bacteria, since their absence leads to a strong decrease or even complete loss of virulence. Our study model, Dickeya dadantii, is a phytopathogenic enterobacteria causing soft rot disease in a wide range of plant species. The opgG or opgH mutants are unable to synthesize OPGs and display a pleiotropic phenotype including a complete loss of virulence. The aim of this study is to better understand the role of OPG in the periplasm. The genetic study of a suppressor mutation in an opgG mutant was carried out. The suppressing mutation is located in the rcsC gene. This mutation, called rcsC2, increases the RcsC phosphatase activity, thus leading to a decrease of RcsB phosphorylation. It restores many phenotypes including virulence on potato tubers but not on chicory leave. The RcsC protein belongs to the RcsCD-RcsB two-component system. In D. dadantii, all the phenotypes observed suggest that the lack of OPG leads to an increased phosphorylation of RcsB by RcsC. The opgGH operon was cloned under the control of an arabinose-inducible promoter which enables us to finely tune the amount of OPG in response to arabinose concentration. We show that the amount of OPG modulates the activity of the RcsCD-RcsB system. We were able to identify new OPG-regulated genes using the system described above. Results suggest that some of these genes could be to be regulated by additional regulators. Thus, OPG are involved in cell signaling pathways through one or several interactions with several two-component systems.</dcterms:abstract>
