<dcterms:abstract xml:lang="fr">Le système nerveux des invertébrés présente des caractéristiques fonctionnelles différentes de celles des vertébrés. Chez la sangsue Hirudo medicinalis, modèle d’étude privilégié en neurobiologie, une réparation efficace de la chaîne nerveuse est possible après lésion. Ce processus physiologique nécessite la mobilisation des cellules microgliales présentes au sein au système nerveux de la sangsue et rappelle certaines phases d’une réaction inflammatoire. Une recherche des effecteurs moléculaires impliqués dans ce recrutement cellulaire a été entreprise. Une protéine homologue au facteur du complément humain C1q, nommée HmC1q, a été caractérisée et a été montrée comme impliquée dans le phénomène d’accumulation précoce de la microglie. En effet, constitutivement exprimée au sein des neurones, HmC1q s’accumule directement au niveau de la lésion du connectif et a été démontrée capable d’augmenter le recrutement d’une sous-population de cellules microgliales en réponse à la lésion. HmC1q se comporte ainsi comme une cytokine à activité chimio-attractante. La production d’une forme recombinante en levure de type Pichia pastoris a permis ensuite de préciser les modalités d’action de ce nouveau facteur chimiotactique, fortement actif au sein du système nerveux de la sangsue. Les expériences de cytométrie en flux, d’immunopréciptation et de chimiotactisme réalisées avec un anticorps anti-gC1qR plaident pour l’existence d’une structure réceptrice homologue au gC1qR des vertébrés sur la sous-population réactive à HmC1q.Par ce travail, il a été mis en évidence, pour la première fois dans le SNC d’un invertébré, l’existence d'une « nouvelle cytokine » HmC1q, ayant une homologie avec la protéine C1q des vertébrés, mais également faisant partie de la vaste famille des cytokines C1q-TNF. Les résultats originaux obtenus suggèrent que HmC1q puisse avoir un rôle-clé dans le dialogue neurone-glie nécessaire au maintien de l’intégrité du système nerveux.</dcterms:abstract>
<dcterms:abstract xml:lang="en">The nervous system of invertebrates has functional characteristics different from those of vertebrates. In the leech Hirudo medicinalis, a well-recognized model in neurobiology, the nerve cord repair might be effective after injury. This physiological process requires the mobilization of microglial cells present in the leech nervous system and seems to mimic some key-phases of an inflammatory reaction.A search of the effector molecules involved in this cell recruitment was undertaken. A protein homologous to human complement factor C1q, named HmC1q, was characterized and was shown to be involved in the early accumulation of microglia. Indeed, constitutively expressed in neurons, HmC1q is directly expressed on the cut end of the connective and, in response to the injury, has been shown to increase the recruitment of a subpopulation of microglial cells. Thus HmC1q behaves as a chemoattractant cytokine. The production of its recombinant form in the yeast Pichia pastoris enabled us to specify the molecular mechanisms by which this new chemoattractant, might be highly active in the leech nervous system. The experiments carried out by flow cytometry, immunopreciptation and in chemotaxis analysis that were performed with an anti-gC1qR, argue for the existence of an human homologous gC1qR-like receptor at the surface the reactive of microglial cell subpopulation reactive to HmC1q. By this work, it was shown, for the first time in the CNS of an invertebrate, the existence of a « new cytokine » HmC1q, homologous to the vertebrate protein C1q, but also belonging to the vast C1q-TNF cytokine family. Finally the present original results suggest that HmC1q may have a key role in neuron-glia dialogue that is essential to maintain the integrity of the nervous system.</dcterms:abstract>
